Mouse Models of Myeloid Malignancies

Faisal Basheer et al.

Summary: Mouse models of human myeloid malignancies are valuable tools for studying the diseases, allowing for the investigation of various aspects such as disease progression, interactions with the microenvironment, pharmacological effects, and co-occurring mutations. Advances in gene-editing technologies like CRISPR-Cas9 have opened up new avenues for the development and study of genetically modified mice and hematopoietic cells.

Interplay between chromosomal alterations and gene mutations shapes the evolutionary trajectory of clonal hematopoiesis

Teng Gao et al.

Summary: This study jointly characterized gene mutations and mCAs in clonal hematopoiesis using peripheral blood sequencing data from cancer patients. Patients with composite genotypes in clonal hematopoiesis were found to have a higher risk of leukemia progression, with mCA identified as an independent risk factor for leukemia development.

Sequential CRISPR gene editing in human iPSCs charts the clonal evolution of myeloid leukemia and identifies early disease targets

Tiansu Wang et al.

Summary: Through the combination of induced pluripotent stem cell (iPSC) and CRISPR-Cas9 technologies, a model of clonal evolution of acute myeloid leukemia (AML) was successfully developed. This model captured distinct premalignant stages and genetic features of primary human MDS and AML, ultimately identifying dysregulation of inflammatory signaling as an early and persistent event in leukemogenesis and a potential therapeutic target.

Summary: The JAK2-V617F mutation affects the self-renewal and differentiation dynamics of HSCs in MPNs patients, helping to reconstruct cancer lineages, with the mutation likely occurring several decades before diagnosis and providing a selective advantage for mutant HSCs.
