Mutation spectrum of NOD2 reveals recessive inheritance as a main driver of Early Onset Crohn's Disease

Julie E. Horowitz et al.

Summary: Inflammatory bowel disease (IBD) is a chronic inflammation of the gastrointestinal tract, including Crohn's disease (CD), ulcerative colitis (UC), or IBD-unclassified. Approximately 25% of all IBD diagnoses are pediatric onset, with 8% potentially influenced by biallelic rare NOD2 variants. Recessive inheritance of NOD2 alleles may be a mechanistic driver of early onset IBD.

CAR directs T cell adaptation to bile acids in the small intestine

Mei Lan Chen et al.

Summary: The study identified the nuclear xenobiotic receptor CAR as a regulator of MDR1 expression in T cells, protecting the small intestine from bile acid toxicity and inflammation. Activation of CAR in T-eff cells infiltrating the small intestine induced large-scale transcriptional reprogramming and promoted the expression of the anti-inflammatory cytokine IL-10.
