A virus-induced conformational switch of STAT1-STAT2 dimers boosts antiviral defenses

Yuxin Wang et al.

Summary: Type I interferons (IFN-I) play a crucial role in protecting against viral infections, with STAT2 being a key component in driving gene expression in response to IFN-I. The phosphorylation of STAT2 at T404 acts as a critical conformational switch, disrupting an inactive STAT1-STAT2 dimer and enhancing the antiviral defense response to IFN-I. Additionally, virus infection-induced activation of IKK-epsilon can directly phosphorylate T404, further facilitating the antiviral defense mechanism.

Genetic Lesions of Type I Interferon Signalling in Human Antiviral Immunity

Christopher J. A. Duncan et al.

Summary: IFN-I is essential in antiviral immunity, with most pathogenic viruses evolving countermeasures against it. Recent discoveries of genetic defects in IFN-I signaling shed light on the importance of IFN-III in humans, enhancing our understanding of the role of the IFN system in the antiviral response.

Auto-antibodies to type I IFNs can underlie adverse reactions to yellow fever live attenuated vaccine

Paul Bastard et al.

Summary: In rare cases, yellow fever virus (YFV) live attenuated vaccine can cause life-threatening disease, particularly in patients with no history of severe viral illness. Some patients with auto-Abs against multiple type I interferons and autosomal recessive IFNAR1 or IFNAR2 deficiency were found to be at risk of developing severe complications after YFV vaccination. Testing for these predispositions could be considered for healthy individuals before YFV vaccination.

Summary: Interleukin-6 receptor antagonists tocilizumab and sarilumab were found to improve outcomes, including survival, in critically ill patients with Covid-19 receiving organ support in ICUs according to an ongoing international trial. The efficacy of these treatments has been confirmed, with patients benefiting from the therapy.

Life-Threatening Influenza, Hemophagocytic Lymphohistiocytosis and Probable Vaccine-Strain Varicella in a Novel Case of Homozygous STAT2 Deficiency

Bishara J. Freij et al.

Summary: This study identified a novel genetic cause of homozygous STAT2 deficiency with unique clinical features, including HLH, severe influenza, and vaccine-strain varicella. Genetic investigation revealed a novel nonsense STAT2 variant that led to a lack of STAT2 protein expression and impaired IFN-I signaling. The occurrence of life-threatening influenza and vaccine-strain varicella in association with STAT2 deficiency highlights the critical role of IFN-I/III immunity in viral infections and vaccine responses.
