Effects of tamoxifen on tendon homeostasis and healing: Considerations for the use of tamoxifen-inducible mouse models

Summary: This study investigated the effects of tamoxifen on tendon homeostasis and healing in mice. The results showed that tamoxifen had no effect on tendon healing in male mice, while it significantly decreased Max load in female repairs. Additionally, tamoxifen treatment led to disruptions in tendon homeostasis in female mice compared to corn oil treatment.

They found that differences in phenotype between Tg(BGLAP-Cre) and Tg(DMP1-Cre) mice with PIK3CA activation may be attributed to differences in recombination rates at non-skeletal sites outside of the bone system. The development of droplet digital PCR assays allowed for the detection and quantification of recombination efficiency in various tissues, providing insights into the potential impact of off-targeting in experiments involving conditional alleles.

Summary: Research has shown that RANKL-stimulated osteoclasts can undergo fission to form osteomorphs, which are involved in the regulation of bone resorption and could be targeted for the treatment of skeletal diseases. Single-cell RNA sequencing revealed that osteomorphs are transcriptionally distinct from osteoclasts and macrophages, expressing non-canonical osteoclast genes associated with bone phenotypes when deleted in mice.

Loss of Smad4 in the scleraxis cell lineage results in postnatal joint contracture

Summary: This study established a novel murine model of joint contracture by targeting Smad4 deletion in the tendon cell lineage. Smad4(ScxCre) mutants showed joint contracture shortly after birth, with severity increasing with age. Disruption of the BMP pathway in Alk3(ScxCre)/Alk6(null) mutants replicated many features of the joint contracture phenotype, suggesting a role for BMP signaling in tendon elongation and extracellular matrix accumulation.
