Keeping Up with RNA-Based Regulation in Bacteria: New Roles for RNA Binding Proteins

Evelyne Ng Kwan Lim et al.

Summary: RNA binding proteins (RBPs) play crucial roles in a variety of regulatory events, ranging from direct gene expression regulation to guiding RNA modification. Over the past decade, the development of high-throughput methods has been instrumental in uncovering new functions of RBPs and elucidating the underlying molecular mechanisms of regulatory events.

Uncovering de novo gene birth in yeast using deep transcriptomics

William R. Blevins et al.

Summary: Recent research has identified 213 de novo originated transcripts in Saccharomyces cerevisiae, with half of them expressed from regions that already contain other genes. These transcripts show similar expression changes in response to stress and some are capable of translating small proteins.

Pathogen-specific antimicrobials engineered de novo through membrane-protein biomimicry

Andrew W. Simonson et al.

Summary: Precision antimicrobials aim to target specific pathogens without harming commensal bacteria in the host, by using a synthetic host defence peptide that mimics key molecular features of the pathogen's channel-forming membrane proteins. By exploiting physical and structural vulnerabilities within the pathogen's cellular envelope, a designed peptide sequence undergoes assembly within the mycolic acid-rich outer membrane of Mycobacterium tuberculosis to specifically kill the pathogen. These biomimetic assemblies enhance antimicrobial activity and antibiotic potency by improving diffusion across the rigid M. tuberculosis envelope.

The largely unexplored biology of small proteins in pro- and eukaryotes

Ruth Steinberg et al.

Summary: Small proteins, typically consisting of less than 50 amino acids in prokaryotes and less than 100 amino acids in eukaryotes, play important roles in cell physiology by stabilizing protein assemblies and modifying the activity of larger proteins. They are often produced under stress conditions and serve as intracellular modifiers for adjusting cell metabolism.

LpxT-Dependent Phosphorylation of Lipid A in Escherichia coli Increases Resistance to Deoxycholate and Enhances Gut Colonization

Xudong Tian et al.

Summary: This study reveals that Escherichia coli has intrinsic resistance to polymyxin B through the simultaneous activation of the PhoPQ and PmrAB regulatory systems under intricate environmental cues. Among various LPS modifications, only EptA and ArnT-dependent decorations are required for polymyxin B resistance, while compromising the innate resistance of E. coli to deoxycholate. The balance between EptA- and LpxT-dependent decorations on lipid A is critical for the lifestyle of E. coli.
