This suggests a link between the transneuronal propagation of alpha-Syn pathology with sensory neuron dysfunction and neuropathic impairment, providing promising avenues for investigating mechanisms underlying pain in PD.

Multiple system atrophy-associated oligodendroglial protein p25α stimulates formation of novel α-synuclein strain with enhanced neurodegenerative potential

Summary: The discovery of a novel alpha-Synuclein strain induced by multiple system atrophy-associated protein p25 alpha indicates a distinct structure and enhanced prodegenerative properties compared to native alpha-Syn. This unique strain has a greater impact on dopamine neurons and results in a shortened lifespan and altered anatomical distribution of inclusion pathology in mice. These findings suggest that oligodendroglial protein p25 alpha plays a significant role in generating a highly aggressive alpha-Syn strain in multiple system atrophy.

Mechanistic basis for receptor-mediated pathological α-synuclein fibril cell-to-cell transmission in Parkinson's disease

Summary: The study reveals that receptors LAG3 and APLP1 on cell surface can preferentially bind pathological alpha-synuclein in the amyloid state to facilitate cell-to-cell transmission in Parkinson's disease progression. Through biophysical experiments, it is found that LAG3 D1 and APLP1 E1 domains commonly use an alkaline surface to bind the acidic C terminus of alpha-synuclein, leading to increased binding affinity. Additionally, phosphorylation at serine 129 enhances the interaction between alpha-synuclein fibrils and the receptors, potentially contributing to the development of PD-like pathology.

α-Synuclein pathology in Parkinson disease activates homeostatic NRF2 anti-oxidant response

Summary: The pathological accumulation of alpha-synuclein impairs redox homeostasis in the nervous system, leading to nuclear NRF2 accumulation and alterations in NRF2-responsive genes. Using a transgenic mouse model, it was observed that neuronal populations with phosphorylated aSyn showed increased nuclear NRF2 and abnormal anti-oxidant and inflammatory gene response in the affected neuraxis. Boosting neuronal anti-oxidant response may be a promising strategy to mitigate neurodegeneration in PD and related diseases.
