Bearing My Heart: The Role of Extracellular Matrix on Cardiac Development, Homeostasis, and Injury Response

Ana Catarina Silva et al.

Summary: The extracellular matrix (ECM) plays a crucial role in heart development and disease, with fibrosis being a major manifestation of many cardiovascular diseases. Effective treatments for managing fibrosis and promoting ECM repair are currently lacking. Understanding ECM composition and function is important for developing more efficient therapies for heart conditions.

Metformin impairs homing ability and efficacy of mesenchymal stem cells for cardiac repair in streptozotocin-induced diabetic cardiomyopathy in rats

Hania Ibrahim Ammar et al.

Summary: The study demonstrates that metformin reduces the efficacy of mesenchymal stem cell therapy for diabetic cardiomyopathy, possibly affecting homing and survival of stem cells. Therefore, future cell replacement therapies in combination with AMPK inhibitors may be more effective for patients with diabetes.

Diverse origins and activation of fibroblasts in cardiac fibrosis

Preetinder K. Aujla et al.

Summary: Cardiac fibroblasts are a heterogeneous cell population and play a crucial role in synthesizing the extracellular matrix (ECM). Various cell types can transform into activated myofibroblasts, leading to accelerated fibrotic response. Multiple molecules stimulate the activation of myofibroblasts, exacerbating fibrosis in the myocardium.

Human umbilical cord-derived mesenchymal stem/stromal cells: a promising candidate for the development of advanced therapy medicinal products

Miryam Mebarki et al.

Summary: UC-MSCs are considered as a promising perspective for treating immune and inflammatory diseases due to their immunomodulatory and anti-inflammatory properties. The classification of UC-MSC-based products as advanced therapy medicinal products in Europe poses challenges for stakeholders in terms of adaptation from cell banks to manufacturing medicines. Despite the potential, the development of UC-MSC-based ATMPs remains a real challenge for both academic institutions and pharmaceutical companies.

Cardiac dysfunction after preeclampsia; an overview of pro- and anti-fibrotic circulating effector molecules

Zenab Mohseni et al.

Summary: Preeclampsia is strongly associated with heart failure later in life, likely due to aberrant cardiac remodeling. Women with PE pregnancies show distinct levels of circulating fibrotic molecules compared to uncomplicated pregnancies during pregnancy and in the postpartum period, which may provide insight into the link between PE and HF.

Interleukin-1 and Systemic Sclerosis: Getting to the Heart of Cardiac Involvement

Giacomo De Luca et al.

Summary: Systemic sclerosis (SSc) is a rare and severe connective tissue disease characterized by endothelial and vascular damage, immune activation, leading to inflammation and fibrosis of skin and internal organs. Cardiac involvement is common in SSc, often asymptomatic initially, with myocardial fibrosis considered the immunopathologic hallmark. Interleukin (IL)-1 and inflammasome play key roles in SSc-heart inflammation, and targeted inhibition of IL-1 is proposed as a potential treatment strategy.
