Active turnover of DNA methylation during cell fate decisions

Aled Parry et al.

Summary: Recent studies have shown that simultaneous activity of de novo methylation and demethylation mechanisms results in rapid DNA methylation turnover. This process may facilitate key lineage decisions and lead to large-scale oscillatory dynamics at gene distal regulatory elements.

Patients with Osteoarthritis and Kashin-Beck Disease Display Distinct CpG Methylation Profiles in the DIO2, GPX3, and TXRND1 Promoter Regions

Rongqiang Zhang et al.

Summary: The study analyzed CpG methylation profiles in the promoter regions of DIO2, GPX3, and TXNRD1 in osteoarthritis (OA) and Kashin-Beck disease (KBD) patients, revealing distinct methylation patterns between the two groups. This provides valuable insights for future studies on the epigenetic regulation mechanisms of selenoprotein genes associated with OA and KBD.

Chromatin architecture reveals cell type-specific target genes for kidney disease risk variants

Aiping Duan et al.

Summary: By conducting a comprehensive multi-omics analysis, we identified regulatory elements associated with functional kidney disease variants and downstream regulated genes. We found that risk variants for renal tumors and chronic kidney disease were enriched in kidney tubule cells, and validated the importance of SLC34A1 and MTX1 in maintaining kidney function through CRISPR/Cas9 genome editing techniques in zebrafish. Our results provide valuable insights into the epigenetic regulation of kidney disease-associated variants in specific cell types.

LC-MALDI-TOF ISD MS analysis is an effective, simple and rapid method of investigation for histones characterization: Application to EBV lymphoblastoid cell lines

Selene De Benedittis et al.

Summary: This study utilized LC-MALDI-TOF analysis to comprehensively describe histone post-translational modifications and variants, demonstrating that MALDI-ISD is an effective method for obtaining global information on histone PTMs and variants in human lymphoblastoid cells. Additionally, this approach allows for rapid acquisition of label-free relative quantitative data for histone variants.

Low-dose hydralazine during gestation reduces renal fibrosis in rodent offspring exposed to maternal high fat diet

Benjamin P. Larkin et al.

Summary: The study found that administration of low-dose Hydralazine to pregnant mice can reduce the occurrence of CKD in offspring of obese mothers by lowering global DNA methylation levels and renal fibrotic markers. This supports the use of low-dose Hydralazine as a demethylating agent to prevent CKD arising in offspring due to maternal HFD consumption.

uhrf1 and dnmt1 Loss Induces an Immune Response in Zebrafish Livers Due to Viral Mimicry by Transposable Elements

Elena Magnani et al.

Summary: The activation of transposable elements (TEs) can lead to cellular damage, where cytoplasmic nucleic acid sensing pathways play a role in not only detecting pathogens but also eliminating cells with inappropriate TE activation. DNA methylation plays a direct role in suppressing certain subclasses of TEs, while the induction of interferon response and tnfa activation in liver cells of mutant organisms mimics gene expression patterns seen in virally infected cells. Antiviral response induced by DNA hypomethylation and TE activation in the liver is mediated by signaling pathways activated by both cytoplasmic double stranded RNA and DNA, with tnfa potentially mediating cell death as a mechanism to eliminate damaged cells.

Dynamic Histone H3 Modifications Regulate Meiosis Initiation via Respiration

Jian Shi et al.

Summary: Meiosis is crucial for genetic stability and diversity in sexual reproduction, and histone H3 acetylation and methylation undergo significant changes during this process. Experimental studies have shown the essential role of H3K18ac in initiating meiosis, revealing a novel regulatory pathway mediated by histone H3 modifications.
