Locking up the AS1411 Aptamer with a Flanking Duplex: Towards an Improved Nucleolin-Targeting

Summary: AS1411-N6, a derivative of the nucleolin-binding aptamer AS1411, adopts a stem-loop structure by adding complementary nucleotides. The formation of G4 sequences decreases compared to AS1411, and G4 formation is promoted by K+. Ligands such as PhenDC3, 360A, and TMPyP4 show promising stabilizer effects on AS1411-N6.

They are non-immunogenic, easier to synthesize and modify, and can bind targets with greater affinity, making them a promising therapeutic candidate for treating various conditions. Active research is ongoing in developing aptamers to target blood coagulation factors, with potential applications in treating cardiovascular diseases, blood disorders, and cancers. Although no aptamers targeting blood coagulation factors have been approved for clinical use, several have shown promise in clinical trials and preclinical studies.

Thrombin-Binding Aptamer with Inversion of Polarity Sites (IPS): Effect on DNAzyme Activity and Anticoagulant Properties

Summary: In this study, thrombin-binding aptamer (TBA) was modified by introducing inversion of polarity sites (IPS) to enhance its activation as a DNAzyme with peroxidase-like activity. The introduction of IPS led to increased peroxidase activity, especially with IPS2 showing the highest activity in the presence of K+ or NH4+ ions. Both IPS1 and IPS2 still retained some anticoagulant activity compared to TBA, and were more resistant to nucleases.
