Organs-on-chips: into the next decade

Lucie A. Low et al.

Summary: OoCs, also known as microphysiological systems or 'tissue chips', have the potential to be informative at multiple stages of drug discovery and development, providing insights into normal human organ function and disease pathophysiology, as well as predicting the safety and efficacy of investigational drugs in humans. The field of OoCs has seen significant advances in recent years, but there are still challenges and opportunities that need to be addressed to fully realize their potential for translational research.

Paracellular and Transcellular Leukocytes Diapedesis Are Divergent but Interconnected Evolutionary Events

Michel-Edwar Mickael et al.

Summary: The infiltration of the endothelial layer of the blood-brain barrier by leukocytes plays a critical role in health and disease. Two migration pathways exist, either following a paracellular or a transcellular route. These two systems share ancient components but have key differences, suggesting they constitute two different mechanical sensing mechanisms.

Development of an In Vitro Airway Epithelial-Endothelial Cell Culture Model on a Flexible Porous Poly(Trimethylene Carbonate) Membrane Based on Calu-3 Airway Epithelial Cells and Lung Microvascular Endothelial Cells

Thijs Pasman et al.

Summary: The study developed a novel in vitro airway epithelial-endothelial cell culture model using Calu-3 human lung epithelial cells and human lung microvascular endothelial cells on flexible porous poly(trimethylene carbonate) membranes. The model demonstrated good cell viability and barrier function, showing potential for lung research.

Regulation of endothelial cell differentiation in embryonic vascular development and its therapeutic potential in cardiovascular diseases

Zi Guo et al.

Summary: During vertebrate development, the cardiovascular system becomes functional earlier than other organs. Endothelial cells play a crucial role in vasculogenesis and angiogenesis, with cellular biology and epigenetics being important factors in these processes. There is growing interest in the role of proteins and signaling molecules in controlling embryonic vascular development, and the transplantation of iPSC-ECs shows promise in treating cardiovascular diseases. However, challenges in maintaining cell viability and function limit the clinical application of iPSC-ECs in regenerative medicine.

Perfusion and endothelialization of engineered tissues with patterned vascular networks

Ian S. Kinstlinger et al.

Summary: The research describes how to fabricate and assemble customizable open-source tissue perfusion chambers, and successfully catheterize tissue constructs; endothelial cells are seeded along the luminal surfaces of the tissue constructs and connected to fluid pumping equipment; enabling various experimental designs, the protocol is expected to promote the adoption and standardization of perfusion tissue culture methods.

Toward three-dimensional in vitro models to study neurovascular unit functions in health and disease

Tara M. Caffrey et al.

Summary: The brain's high metabolic demands require an efficient vascular system to supply nutrients and oxygen, which is regulated by the neurovascular unit composed of neurons, glial and vascular cells. Changes in neurovascular unit functions in neurodegenerative diseases can impair neurovascular coupling, blood-brain barrier permeability, cerebral blood flow, and waste clearance. Advanced physiologically-relevant human models of the neurovascular unit are being developed using stem-cell derived organoids and vascularized organoids to better understand disease processes and potential therapeutic strategies. Continued innovation in engineering and design is necessary to create more complex models for studying neurovascular unit functionality.
