A Randomized Dose-Escalating Phase I Trial of a Replication-Deficient Lymphocytic Choriomeningitis Virus Vector-Based Vaccine Against Human Cytomegalovirus

Michael Schwendinger et al.

Summary: This study describes the first-in-man study of a replication-deficient arenavirus-based vector, HB-101, expressing cytomegalovirus gB and pp65. Consecutive administrations of HB-101 were well tolerated and elicited gB-binding and neutralizing antibodies as well as cytomegalovirus-specific CD8 T-cell responses in healthy volunteers.

Functional Evaluation and Genetic Evolution of Human T-Cell Responses After Vaccination With a Conditionally Replication-Defective Cytomegalovirus Vaccine

Kara S. Cox et al.

Summary: The study analyzed vaccine-induced T cells for memory phenotype, antigen specificity, cytokine profiles, and cytolytic potential using multicolor flow cytometry. The results showed that vaccination elicited polyfunctional CD4 and CD8 T cells with predominantly effector phenotype responding to 2 dominant antigens, pp65 and IE1. Analysis of T-cell receptor repertoires demonstrated polyclonal expansion of antigen-specific T cells after vaccination.

Summary: This study reviewed the potential mechanisms of CMV in causing stillbirth, preterm birth, and IUGR, identifying impairments in trophoblast cell differentiation and function, invasiveness, and signaling pathway regulation, as well as induction of apoptosis. Challenges remain in finding suitable in vivo animal models for further research in this area.

Regulation of Latency and Reactivation by Human Cytomegalovirus miRNAs

Nicole L. Diggins et al.

Summary: Human cytomegalovirus (HCMV) encodes 22 mature microRNAs that regulate various cellular processes, including vesicular trafficking, cell cycle progression, apoptosis, and immune evasion. Recent evidence suggests a critical role for HCMV microRNAs in mediating latency in CD34(+) hematopoietic progenitor cells by modulating cellular signaling pathways. Additionally, these microRNAs can interact with viral proteins to influence host cell functions.

Humanized Mice for the Evaluation of Novel HIV-1 Therapies

Shawn Abeynaike et al.

Summary: While HIV-1 infection can now be managed with lifelong treatment, complete eradication remains a challenge due to the virus's ability to remain in dormant state in tissue reservoirs and its high mutation rate. Developing a suitable animal model is critical for evaluating pathogenesis and therapeutic approaches for HIV-1 cure strategies.

Common Polymorphisms in the Glycoproteins of Human Cytomegalovirus and Associated Strain-Specific Immunity

Hsuan-Yuan Wang et al.

Summary: HCMV is one of the most prevalent viruses globally, causing common morbidity and mortality for immunocompromised individuals. Clinical observations suggest that mixed strain infections are common and may lead to more severe disease progression. This genetic variation in glycoproteins could be an important resource for future efforts to combat HCMV infections.

Prophylactic antigen-specific T-cells targeting seven viral and fungal pathogens after allogeneic haemopoietic stem cell transplant

David Jonathan Gottlieb et al.

Summary: Adoptive immunotherapy using donor-derived antigen-specific T-cells can effectively prevent and treat infections after allogeneic haemopoietic stem cell transplant. The study showed that rapid reconstitution of immunity to a broad range of viral and fungal infections can be achieved using a multi-pathogen-specific T-cell product, but precautions should be taken to control the development of graft-versus-host disease.

Summary: Individuals with compromised immune systems are at the highest risk for complications from CMV infection, including those living with HIV/AIDS, transplant recipients, and fetuses. Despite significant progress in prevention, diagnostics, and treatment, there are challenges in optimal control of CMV infection and disease in transplant settings.
