Synthesis, docking studies, and pharmacological evaluation of 2-hydroxypropyl-4-arylpiperazine derivatives as serotoninergic ligands

Summary: A series of new norbornene and exo-N-hydroxy-7-oxabicyclo[2.2.1]hept-5-ene-2,3-dicarboximide derivatives were synthesized and evaluated for their affinities to different 5-HT receptors to identify selective ligands. Compounds with critical structural elements showed high specificity and affinity, with some selected for further in vivo assays. Molecular docking studies were conducted to rationalize the results, indicating 3e, 4j, and 4n as the most active and promising derivatives for the serotonin receptor considered in the study.
