Summary: Osteoporosis is a significant cause of morbidity, especially in post-menopausal women and older men, but the search for an effective treatment remains a challenge. Despite the progress in developing nano-based materials as drug delivery systems against OP, further clinical investigations and evaluation are necessary. Some main challenges include low reproducibility in manufacturing processes, unsatisfactory drug-loading capacity, uncontrollable release kinetics, and low delivery efficiency, which limit the clinical application of these materials.

An oligopeptide/aptamer-conjugated dendrimer-based nanocarrier for dual-targeting delivery to bone

Summary: A new dual-targeting nanocarrier, CH6-PAMAM-C11, was synthesized by covalently modifying G4.0 PAMAM dendrimer with C11 peptide and CH6 aptamer, showing successful accumulation in bone cells, mineralized areas, and tissues. Both in vitro and in vivo experiments confirmed the efficient bone targeting ability of CH6-PAMAM-C11, suggesting potential for drug delivery to bone tissues for osteoporosis treatment.
