Explored mechanisms by CSCs to overcome different therapeutic strategies highlight their plasticity as an insidious trait responsible for their adaptation/escape, with novel CSC-specific approaches showing promising preclinical and clinical applications.

Beyond the Warburg Effect: Oxidative and Glycolytic Phenotypes Coexist within the Metabolic Heterogeneity of Glioblastoma

Summary: The study characterized the basal bioenergetic metabolism and antiproliferative potential of metabolic drugs in different glioma stem cells, finding that GBM27 was more resistant to treatments compared to GBM18 and GBM38. Combining radiomimetic bleomycin with specific metabolic drugs exhibited therapeutically relevant synergistic effects. Metformin and dichloroacetate altered the metabolism of treated cells, while diazo-5-oxo-L-norleucine consistently decreased total ATP production. More research is needed for a better understanding of glioblastoma metabolism and effective metabolic therapy strategies.

Neuronavigation-guided focused ultrasound for transcranial blood-brain barrier opening and immunostimulation in brain tumors

Summary: Focused ultrasound (FUS) with microbubbles can transiently open the blood-brain barrier (BBB) to increase therapeutic agent penetration, benefiting recurrent glioblastoma (rGBM) treatment. A pilot trial using a neuronavigation combined with a manually operated frameless FUS system for rGBM patients established safety and feasibility, with dose-dependent BBB-opening effect observed. Additionally, FUS-induced immune modulation was demonstrated to convert the immunosuppressive tumor microenvironment into an immunostimulatory one at a higher but safe dosage.

The Many Facets of Therapy Resistance and Tumor Recurrence in Glioblastoma

Summary: GBM is the most lethal type of primary brain cancer, with therapy resistance attributed to both intrinsic factors like tumor heterogeneity and altered metabolomics, as well as extrinsic factors like hypoxia and an immune-suppressive tumor microenvironment. The escape mechanisms of tumor cells during therapy also contribute to treatment failure in GBM.

An insight into the structure of 5-spiro aromatic derivatives of imidazolidine-2,4-dione, a new group of very potent inhibitors of tumor multidrug resistance in T-lymphoma cells

Summary: A series of newly synthesized compounds were explored for their potential as potent inhibitors of cancer multidrug resistance efflux pumps. Particularly, the spirofluorene derivatives with amine alkyl substituents at position 1 showed the strongest P-glycoprotein inhibitory activity in MDR T-lymphoma cells, with compounds 7 and 12 being 100-fold more potent than verapamil. Crystallography-supported pharmacophore-based SAR analysis suggested specific structural properties contributing to this excellent cancer MDR-inhibitory action.

Glioblastomas acquire myeloid-affiliated transcriptional programs via epigenetic immunoediting to elicit immune evasion

Summary: The study elucidated the mechanisms by which hypoxic lncRNA derived from glioma stem cells (GSC) drive glioma progression, involving the Linc01060 gene, exosomes containing it, and proteins regulating the gene. Transfer of exosomes from GSCs to glioma cells increased Linc01060 levels and activated prooncogenic signaling pathways. Inhibition of Linc01060-containing exosomes or targeting the Linc01060/MZEI/c-Myc/HIF1 alpha axis could be an effective therapeutic strategy in glioma.
