Summary: This study compared the molecular markers distribution and expression levels in human fetal retina and human embryonic stem cell (hESC)-derived retinal tissue to evaluate the potential clinical application of hESC-retinal tissue for vision restoration. Results revealed a high correlation in gene expression and histological profiles between fetal retina and hESC-derived retinal tissue. Summary: Mutations in the CRX gene lead to distinct retinopathy phenotypes and early-onset vision impairment. Using iPSCs from patients with CRX mutations, researchers established in vitro models of CRX-LCA and found that AAV-mediated CRX gene augmentation therapy partially restored defective photoreceptor maturation and opsins expression. This study provides proof-of-concept for developing gene therapy for dominant CRX-LCA and other CRX retinopathies.

Summary: The study demonstrated that PM2.5 exposure affected the formation of neural retina derived from hEROs, leading to reduced proliferation and increased apoptosis. Transcriptome analysis showed that PM2.5 treatment was linked to MAPK and PI3K/Akt pathways, and reduced the level of FGFs, particularly FGF8 and FGF10, suggesting potential inhibition of proliferation and increase of apoptosis during early human retinal development.

Optimized Adeno-Associated Virus Vectors for Efficient Transduction of Human Retinal Organoids

Manuela Voelkner et al.

Summary: rAAV vectors are widely used in gene delivery in the retina, showing safety and efficacy in treating inherited retinal dystrophies with limitations in transduction efficiency. Novel variants with modified capsid sequences have been engineered to improve this efficiency. Testing on hiPSC-derived human retinal organoids showed promising results with AAV9.NN achieving the highest efficiency in transduction. Using human organoids provides a valuable platform for evaluating novel AAV vectors for gene therapy applications in the retina.
