Proteostasis of Islet Amyloid Polypeptide: A Molecular Perspective of Risk Factors and Protective Strategies for Type II Diabetes

Summary: The possible link between hIAPP accumulation and beta-cell death in diabetic patients has sparked a lot of research on amyloid structures and aggregation pathways. Recent studies have highlighted the importance of early oligomeric intermediates and their interactions with lipid membranes, pH, insulin, and zinc in the mechanism of hIAPP aggregation. Challenges in characterizing the aggregation process have led to the development of various biophysical and chemical approaches, with studies suggesting that restoring hIAPP proteostasis could be a promising strategy for therapy.

Resistance of nepetin and its analogs on the fibril formation of human islet amyloid polypeptide

Summary: The study found that four flavones effectively inhibited the aggregation of hIAPP and A beta protein, dispersing the mature fibrils into monomers. These compounds interacted with the peptides through hydrophobic and hydrogen bonding interactions. Additionally, the flavones exhibited better inhibitory effects on hIAPP compared to A beta protein.

Inhibition behavior of Sennoside A and Sennoside C on amyloid fibrillation of human lysozyme and its possible mechanism

Summary: In this study, the inhibitory effects of Sennoside A and Sennoside C on amyloid fibrillation were evaluated using human lysozyme as a model. Both compounds were found to inhibit the fibrillation of lysozyme in a dose-dependent manner, with interactions mainly through hydrogen bonds, vander-Waals forces, and hydrophobic interactions. Transmission electron microscopy and atomic force microscopy confirmed the reduction in fibrillation, supporting the conclusion that both Sennoside A and Sennoside C can significantly inhibit amyloid fibrillation of lysozyme.
