Protein kinase R and the integrated stress response drive immunopathology caused by mutations in the RNA deaminase ADAR1

Summary: ADAR1 is a negative regulator of the RNA sensor MDA5 and loss of ADAR1 function can cause immune diseases. Research in a mouse model showed that a small-molecule inhibitor targeting the eIF2 alpha kinase PKR and ISR can prevent immunopathology and rescue mice from death. This highlights PKR and ISR as central components of immunopathology and potential therapeutic targets for human diseases associated with the ADAR1-MDA5 axis.

Mutations in the adenosine deaminase ADAR1 that prevent endogenous Z-RNA binding induce Aicardi-Goutieres-syndrome-like encephalopathy

Summary: Mutations in ADAR1 p150, including point mutations in the Z alpha domain, are associated with Aicardi-Goutieres syndrome (AGS). Mutation of W197 in Z alpha abolished Z-RNA binding and reduced RNA editing. Adar1(W197A/W197A) mice showed severe growth retardation, broad expression of interferon-stimulated genes, and abnormal development of multiple organs. Deletion of the MDA5 sensor ameliorated these abnormalities, suggesting that ADAR1 plays a crucial role in preventing inappropriate inflammation.
