24S-hydroxycholesterol: Cellular effects and variations in brain diseases

Alejandro O. Sodero

Summary: The production and elimination of 24S-HC in the brain is discussed, followed by a focus on how this oxysterol affects various aspects of cellular function and disease progression at the cellular level.

Astrocytic ApoE reprograms neuronal cholesterol metabolism and histone-acetylation-mediated memory

Xiaohui Li et al.

Summary: This study demonstrates that astrocytes interact with neurons by delivering microRNAs that modulate neuronal cholesterol biosynthesis genes, affecting neuronal metabolism and epigenetic regulation. The findings reveal a novel mechanism through which astrocytes regulate brain connectivity and function by mediating neuronal epigenetic signals.

Sterol auto-oxidation adversely affects human motor neuron viability and is a neuropathological feature of amyotrophic lateral sclerosis

James C. Dodge et al.

Summary: Aberrant cholesterol homeostasis is implicated in the pathogenesis of ALS, with increased cholesterol oxidation found in the spinal cord of ALS models and patients. Auto-oxidized sterols were toxic to human motor neurons, suggesting that unregulated sterol oxidation may contribute to neuronal death in ALS. Developing therapeutic strategies to restore cholesterol homeostasis in the spinal cord could potentially lead to ALS treatment.

ApoE4 Impairs Neuron-Astrocyte Coupling of Fatty Acid Metabolism

Guoyuan Qi et al.

Summary: This study reveals the detrimental effects of ApoE4 on fatty acid metabolism in neurons and astrocytes, leading to lipid accumulation and energy imbalance. ApoE4 decreases the transport efficiency of neuronal lipids, compromising metabolic and synaptic support from astrocytes to neurons.

APOE4 disrupts intracellular lipid homeostasis in human iPSC-derived glia

Grzegorz Sienski et al.

Summary: The APOE gene's E4 allele is a genetic risk factor for cardiovascular diseases and Alzheimer's disease, but its mechanism of action is poorly understood. A study found that APOE4 disrupted cellular lipidomes, leading to increased unsaturation of fatty acids and intracellular lipid droplet accumulation. Supplementation with choline restored the cellular lipidome in APOE4-expressing cells, suggesting that manipulating lipid metabolism could be a therapeutic approach for individuals with the APOE4 allele.

Atxn2-CAG100-KnockIn mouse spinal cord shows progressive TDP43 pathology associated with cholesterol biosynthesis suppression

Julia Canet-Pons et al.

Summary: Large polyglutamine expansions in ATXN2 cause multi-system nervous atrophy in SCA2, while intermediate size expansions carry a risk for selective motor neuron degeneration in ALS. Depletion of ATXN2 can prevent disease progression in ALS. The study of an authentic SCA2 mouse model reveals molecular mechanisms in spinal cord pathology, overlapping with known ALS features. Identifying new targets and potential biomarkers may lead to novel neuroprotective therapies.

TDP-43 mediates SREBF2-regulated gene expression required for oligodendrocyte myelination

Wan Yun Ho et al.

Summary: The protein TDP-43 influences cholesterol metabolism in oligodendrocytes by directly binding to mRNA of key regulators. Depletion of TDP-43 leads to reduced expression of SREBF2 and LDLR, resulting in lower cholesterol levels. Reintroducing SREBF2 or LDLR can reverse the effects of TDP-43 depletion on cholesterol levels.

Childhood amyotrophic lateral sclerosis caused by excess sphingolipid synthesis

Payam Mohassel et al.

Summary: This study identifies SPTLC1 variants causing excessive sphingolipid biosynthesis as a monogenic form of ALS. Disruption of normal SPT regulation by ORMDL proteins leads to uncontrolled SPT activity and elevated levels of canonical SPT products. Custom-designed small interfering RNAs that selectively target the SPTLC1 ALS allele for degradation offer a potential therapeutic approach to normalize sphingolipid levels.

Neuronal ApoE upregulates MHC-I expression to drive selective neurodegeneration in Alzheimer's disease

Kelly A. Zalocusky et al.

Summary: Selective neurodegeneration is a critical causal factor in Alzheimer's disease, and a study has demonstrated a causal relationship between neuronal ApoE expression and MHC-I expression leading to tau pathology and selective neurodegeneration. The correlation between ApoE expression and immune response pathways suggests a mechanism for the vulnerability of certain neurons in Alzheimer's disease.
