Targeting MCL1 to induce mitophagy is a potential therapeutic strategy for Alzheimer disease

Xufeng Cen et al.

Summary: This study identified a small molecule compound, UMI-77, as a safe and effective inducer of mitophagy to improve cognitive decline in an AD mouse model. It sheds light on novel mechanisms of mitophagy, demonstrates MCL1 as a receptor that can be targeted to induce mitophagy, and identifies MCL1 as a drug target for therapeutic intervention in AD.

Targeting transcription of MCL-1 sensitizes HER2-amplified breast cancers to HER2 inhibitors

Konstantinos Floros et al.

Summary: HER2 inhibitors are less effective in breast cancer due to low levels of MCL-1, but combining them with CDK inhibitors can sensitize HER2 inhibitors and lead to effective treatment. This combination therapy shows promising results in preclinical models and may offer a chemotherapy-free therapeutic strategy for HER2-amplified breast cancer patients.

Persistent hepatocyte apoptosis promotes tumorigenesis from diethylnitrosamine-transformed hepatocytes through increased oxidative stress, independent of compensatory liver regeneration

Yasutoshi Nozaki et al.

Summary: This study found that enhanced tumor formation from DEN-transformed hepatocytes by persistent hepatocyte apoptosis is mediated by increased oxidative stress, independent of compensatory liver regeneration. Controlling oxidative stress may play a critical role in suppressing hepatocarcinogenesis for patients with livers harboring transformed cells.

Requirement for antiapoptotic MCL-1 during early erythropoiesis

Meghan E. Turnis et al.

Summary: Deletion of erythroid-specific Mcl1 results in embryonic lethality due to severe anemia caused by lack of mature red blood cells. MCL-1 is required only during early definitive erythropoiesis, with developing erythrocytes becoming MCL-1 independent and upregulating BCL-xL expression during later stages.

Cell-penetrating Alphabody protein scaffolds for intracellular drug targeting

Erwin Pannecoucke et al.

Summary: This study demonstrates the engineering of the Alphabody scaffold into a cell-penetrating protein antagonist against intracellular cancer target MCL-1, with the addition of an albumin-binding moiety to extend serum half-life. The engineered Alphabody reduced tumor burden in mouse tumor xenografts based on myeloma cell lines, providing a proof of concept for using Alphabodies against intracellular disease mediators.
