Summary: The rs599839 A>G variant in the CELSR2-PSRC1-SORT1 gene cluster is associated with protection against dyslipidemia and cardiovascular events in NAFLD patients, but may increase the risk of HCC development by modulating SORT1 and PSRC1 expressions. This variant has been linked to reduced LDL levels, carotid intima-media thickness, and hypertension, as well as an increased risk of HCC independent of fibrosis severity and poor prognosis. Hepatic PSRC1, SORT1, and CELSR2 expressions were found to be elevated in NAFLD patients carrying the rs599839 variant, with SORT1 negatively correlating with lipid levels and PSRC1 positively correlating with genes involved in cell proliferation. PSRC1 over-expression was associated with more aggressive HCC development in TCGA samples.
