Efficacy of Single-Dose Primaquine With Artemisinin Combination Therapy on Plasmodium falciparum Gametocytes and Transmission: An Individual Patient Meta-Analysis

Kasia Stepniewska et al.

Summary: A meta-analysis of individual patients was conducted to assess the gametocytocidal and transmission-blocking activities of single-dose primaquine. The results showed that primaquine, when administered with artemisinin-based combination therapies, can effectively reduce the gametocyte carriage rate after 7 and 14 days, especially in patients with existing gametocytes at the start of treatment. Furthermore, the combination of primaquine and artemether-lumefantrine showed a faster decline in gametocyte carriage compared to the combination with dihydroartemisinin-piperaquine. Additionally, the addition of 0.25 mg/kg primaquine can nearly completely prevent transmission to mosquitoes.

Clinical and In Vitro Resistance of Plasmodium falciparum to Artesunate- Amodiaquine in Cambodia

Melissa Mairet-Khedim et al.

Summary: The efficacy of artesunate-amodiaquine for uncomplicated falciparum malaria in Cambodia was found to be inadequate due to amodiaquine-resistant P. falciparum. The study identified that amodiaquine resistance was not associated with previously identified molecular markers.

Triple Artemisinin-Based Combination Therapies for Malaria - A New Paradigm?

Rob W. van der Pluijm et al.

Summary: Recent progress in the fight against malaria is at risk due to the emergence and spread of artemisinin and partner drug resistance, particularly in the Greater Mekong Subregion. New drugs are not expected in the near future, making it crucial to utilize triple artemisinin-based combination therapies to combat falciparum malaria effectively. Deployment of such therapies beyond the GMS could potentially help prevent global resistance to currently available drugs.

Endectocides as a complementary intervention in the malaria control program: a systematic review

Fereshteh Ghahvechi Khaligh et al.

Summary: Systemic insecticides and endectocides, especially ivermectin, show potential in controlling malaria transmission by targeting Anopheles mosquitoes. Studies focused on Anopheles gambiae complex species and used animals and humans as target hosts for administering these drugs. Laboratory and field research indicate that these drugs could serve as novel tools for reducing Plasmodium parasite transmission in malaria control efforts.

Deploying triple artemisinin-based combination therapy (TACT) for malaria treatment in Africa: ethical and practical considerations

Paulina Tindana et al.

Summary: Malaria is a significant cause of morbidity and mortality in African children, with artemisinin-based combination therapy as the first-line treatment. However, drug resistance in Southeast Asia poses a threat, and the development of triple artemisinin-based combination therapy is seen as a potential solution with ethical and practical considerations.

Transmission of Artemisinin-Resistant Malaria Parasites to Mosquitoes under Antimalarial Drug Pressure

Kathrin Witmer et al.

Summary: Resistance to artemisinin-based combination therapy (ACT) in the Plasmodium falciparum parasite threatens recent progress in reducing global malaria deaths. Under artemisinin drug pressure, resistant parasites have a selective advantage in transmission, potentially leading to the spread of multidrug-resistant malaria beyond Southeast Asia.
