Designing gene drives to limit spillover to non-target populations

Gili Greenbaum et al.

Summary: CRISPR-based gene drive technology shows promise but also carries risks of unintentional spillovers. Differential targeting to limit spillovers to non-target populations is feasible with careful design and low migration rates, but challenging under high migration rates. Safeguards such as differential targeting should be considered when applying CRISPR-based gene drives in order to minimize ecological disruptions.

Invasion and maintenance of meiotic drivers in populations of ascomycete fungi

Ivain Martinossi-Allibert et al.

Summary: Meiotic drivers (MDs) are selfish genetic elements that can spread in a population due to their transmission advantage, even if they impose fitness costs on their host organisms. The ability of an MD to invade and coexist in a population depends on the host organism's biology and the specific fitness effects of the driving allele on the host.

Systematic identification of plausible pathways to potential harm via problem formulation for investigational releases of a population suppression gene drive to control the human malaria vector Anopheles gambiae in West Africa

John B. Connolly et al.

Summary: Population suppression gene drive using CRISPR-Cas9 technology has shown promising results in controlling malaria vectors. The study identified potential risks and developed risk hypotheses for simulated field releases of a transgene in West Africa, emphasizing the importance of further research and modeling to evaluate the potential harms and benefits of this approach.

A transgenic female killing system for the genetic control of Drosophila suzukii

Marc F. Schetelig et al.

Summary: A new conditional female-killing (FK) strategy based on the tetracycline-off system was developed to combat the spotted wing Drosophila. Through testing different genetic constructs and transgenic strains, a construct that achieved female-specific lethality in the absence of tetracycline was identified, effectively controlling the population of female pests. Suggestions for potential improvements to the gene constructs and the use of FK strains in a sterile insect technique (SIT) program are discussed.
