Epigenetic mechanisms regulating COVID-19 infection

Sarantis Chlamydas et al.

Summary: The COVID-19 outbreak originating in Wuhan, China poses a significant global threat to human health. The highly contagious SARS-CoV-2 infection and transmission, along with age-related factors and epigenetic changes, play key roles in the pathophysiology of the virus. Epigenetic research has shown that factors such as global DNA methylation and host cell epigenetic modulation following infection may influence host susceptibility to viral infection.

Susceptibility to COVID-19 in populations with health disparities: Posited involvement of mitochondrial disorder, socioeconomic stress, and pollutants

Yunyi Yao et al.

Summary: The novel betacoronavirus SARS-CoV-2 causing COVID-19 is discussed in relation to mitochondrial dysfunction and the potential effects of environmental and endocrine-disrupting chemicals on mitochondria. The virus can disrupt mitochondrial function during infection, affecting organelle functions and suggesting the need to monitor and protect mitochondria during COVID-19 treatment. Mitochondrial metabolomic analysis may offer new indicators for COVID-19 prognosis, and understanding the role of mitochondria in SARS-CoV-2 infection could improve intervention therapies and protect mitochondrial disease patients and those exposed to stressors like poor nutrition and environmental chemicals.

MicroRNAs and SARS-CoV-2 life cycle, pathogenesis, and mutations: biomarkers or therapeutic agents?

Farshad Abedi et al.

Summary: Proposed therapies and antiviral drugs have so far been ineffective in curing COVID-19 patients, but there are vaccine candidates seeking emergency use authorization. Recent studies have highlighted the role of miRNAs in the pathophysiology of COVID-19, suggesting their potential as biomarkers or antiviral agents in therapy.

Epigenetic Evolution of ACE2 and IL-6 Genes: Non-Canonical Interferon-Stimulated Genes Correlate to COVID-19 Susceptibility in Vertebrates

Eric R. Sang et al.

Summary: The current COVID-19 pandemic has highlighted the substantial differences in susceptibility and disease progression in individuals of different ages, genders, and pre-existing comorbidities. Host factors undergo epigenetic regulation, with key genes like ACE2 and IL-6 showing active epigenetic evolution that may serve as biomarkers for predicting COVID-19 susceptibility. The epigenetic properties of ACE2 and IL-6 genes suggest their unconventional responses to inflammatory and interferon signaling, potentially explaining the disparities in COVID-19 outcomes among different subgroups.

Structural Variability, Expression Profile, and Pharmacogenetic Properties of TMPRSS2 Gene as a Potential Target for COVID-19 Therapy

Aleksei Zarubin et al.

Summary: The human serine protease serine 2 TMPRSS2 plays a role in priming proteins of SARS-CoV-2 and is a possible target for COVID-19 therapy. Functional missense mutations in the TMPRSS2 gene are found in many human populations, with higher frequencies in populations of Asian origin compared to European populations. TMPRSS2 exhibits tissue-specific expression, particularly in alveolar cells, and has interactions with SARS-CoV-2.

Micro-RNAs in the regulation of immune response against SARS CoV-2 and other viral infections

Tareq Abu-Izneid et al.

Summary: miRNAs play key roles in regulating gene expression, especially in viral infections, making them potential targets for novel therapeutic strategies. Modulation of immune-pathogenesis responses by miRNAs can either enhance or inhibit virus replication, highlighting their importance in viral infection treatment.

Summary: The virus can enhance its survival by manipulating key signaling pathways in host cells, leading to more complex host responses to the virus, which may result in pathological abnormalities in the lungs and other organs, as well as an increased risk of infection for patients with comorbidities.

Role of Genetic Variants and Gene Expression in the Susceptibility and Severity of COVID-19

Sarita Choudhary et al.

Summary: During the COVID-19 pandemic, understanding genetic risk factors for susceptibility to the disease is essential. Genes like ACE2, TMPRSS2, and DPP4 play key roles in determining disease severity. Research on genetic variants influencing respiratory infections can help identify potential drug targets for COVID-19 patients.

Targeting androgen regulation of TMPRSS2 and ACE2 as a therapeutic strategy to combat COVID-19

Qu Deng et al.

Summary: The study suggests a potential role of androgen in SARS-CoV-2 infection by regulating ACE2 and TMPRSS2 transcription. Camostat, a TMPRSS2 inhibitor, blocks the cleavage of SARS-CoV-2 Spike protein, providing evidence of TMPRSS2's direct role in viral fusion to host cells. Androgen-deprivation, anti-androgens, or camostat attenuate SARS-CoV-2 cellular entry, highlighting the importance of TMPRSS2 inhibitors and androgen-deprivation therapy in preventing COVID-19 progression.
