Comparison of Two 16S rRNA Primers (V3-V4 and V4-V5) for Studies of Arctic Microbial Communities

Summary: Microbial communities in the Arctic Ocean are still poorly understood, but recent studies have shown the presence of unique microbial community members adapted to extreme environmental conditions. In comparing two commonly used primer sets (V3-V4 and V4-V5) for studying microbial communities in Arctic marine habitats, it was found that both sets represent the total microbial community composition effectively down to the genus rank, with the V4-V5 primer set providing coverage of both bacterial and archaeal domains for a more comprehensive study.
