Autophagy Triggers Tamoxifen Resistance in Human Breast Cancer Cells by Preventing Drug-Induced Lysosomal Damage

Summary: Endocrine therapy with tamoxifen is the standard treatment for estrogen receptor-positive breast cancer, but drug resistance affects about 30% of patients. Autophagy has been proposed as a key player in drug resistance, with tamoxifen affecting lysosome integrity. The study found that activation of autophagy and overexpression of iron-binding proteins protect lysosomes, leading to drug resistance in breast cancer cells. The results suggest that screening for iron-binding proteins may help identify patients at risk for developing drug resistance.
