Serial Circulating Tumor DNA in Predicting and Monitoring the Effect of Neoadjuvant Chemoradiotherapy in Patients with Rectal Cancer: A Prospective Multicenter Study

Jiaolin Zhou et al.

Summary: The study showed that ctDNA can serve as a real-time monitoring indicator that accurately reflects tumor burden. The median variant allele frequency (VAF) of mutations in baseline ctDNA is a strong independent predictor of metastasis-free survival (MFS).

Tumor mutation burden in blood predicts benefit from neoadjuvant chemo/radiotherapy in locally advanced rectal cancer

Dengbo Ji et al.

Summary: The study found that bTMB levels are correlated with RFS in LARC patients. Patients with decreasing TMB after nCRT tend to have longer RFS, while higher bTMB levels after surgery are negatively correlated with RFS. Serum cytokines were significantly higher in the high bTMB group, and clonal evolution analysis showed shared mutations in pre- and post-nCRT ctDNAs in most cases.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

The PRISMA 2020 statement: an updated guideline for reporting systematic reviews

Matthew J. Page et al.

Summary: The PRISMA statement was designed to help systematic reviewers transparently report the purpose, methods, and findings of their reviews. The updated PRISMA 2020 statement includes new reporting guidance, a 27-item checklist, an abstract checklist, and revised flow diagrams for reviews.

Circulating Tumor DNA Predicts Pathologic and Clinical Outcomes Following Neoadjuvant Chemoradiation and Surgery for Patients With Locally Advanced Rectal Cancer

Susan G. R. McDuff et al.

Summary: This study found that in patients treated with neoadjuvant chemoradiation for LARC, patients with undetectable preoperative ctDNA were more likely to have a favorable surgical outcome, while detectable postoperative ctDNA was associated with poorer recurrence-free survival.
