Mfd regulates RNA polymerase association with hard-to-transcribe regions in vivo, especially those with structured RNAs

Summary: Research has shown that Mfd can influence transcription by repressing the association of bacterial RNA polymerase at multiple chromosomal regions, especially in areas with highly structured regulatory RNAs. The function of Mfd extends beyond DNA repair, including transcription regulation and catalyzing mutations in certain toxin genes to promote the evolution of TA systems and other regions containing strong RNA secondary structures.

Summary: Transcription-coupled repair (TCR), a sub-pathway of nucleotide excision repair (NER), removes lesions from the template-strand that stall RNA polymerase elongation complexes. Cryo-electron microscopy was used to visualize Mfd engaging with a stalled EC and dislodging the RNAP, showing how Mfd discriminates between paused and stalled ECs and initiates TCR through a complex mechanism.
