Pyronaridine: An update of its pharmacological activities and mechanisms of action

Christian Bailly

Summary: PYR is a potent erythrocytic schizonticide with antimalarial activity against drug-resistant Plasmodium, also showing antitumor effects and efficacy against other parasitic diseases. Its mechanisms of action involve inhibition of hemozoin formation, interference with DNA metabolism, potential immune modulation, and implication in mitochondrial oxidative pathways. The diverse health benefits of pyronaridine are highlighted through its various biological actions.

Defining the Antimalarial Activity of Cipargamin in Healthy Volunteers Experimentally Infected with Blood-Stage Plasmodium falciparum

James S. McCarthy et al.

Summary: The new antimalarial compound cipargamin showed antimalarial activity in healthy volunteers infected with blood-stage Plasmodium falciparum, although some subjects experienced serious liver function changes during treatment. Further evaluation of the compound's hepatic safety profile is needed despite the promising antimalarial activity observed.

Repurposing of antitumor drug candidate Quisinostat lead to novel spirocyclic antimalarial agents

Ruoxi Li et al.

Summary: Two novel compounds, 8 and 27, designed and synthesized based on Quisinostat, demonstrated promising antimalarial activities against wild-type and multi-drug resistant parasites with reduced cytotoxicity and good metabolic stability. They also showed certain efficacy in a rodent malaria model, representing prospective prototypes for further antimalarial drug research.

Chemoprotective Antimalarial Activity of P218 against Plasmodium falciparum: A Randomized, Placebo-Controlled Volunteer Infection Study

Mohamed Farouk Chughlay et al.

Summary: P218 demonstrated excellent chemoprotective efficacy against P. falciparum, effectively inhibiting the parasite. Clinical trials showed that P218 had good safety and tolerability in participants, with mild, transient, and self-limiting adverse events. Furthermore, P218's pharmacokinetics were similar in participants with and without induced infection.

Efficient Treatment of Experimental Cerebral Malaria by an Artemisone-SMEDDS System: Impact of Application Route and Dosing Frequency

Johanna Zech et al.

Summary: The study demonstrates that using SMEDDS can enhance the efficacy of ART in treating malaria, with better results achieved by splitting the dose and choosing the appropriate route of administration. Nasal treatment was found to be the most effective, followed by oral treatment, while intraperitoneal injection was the least effective route.

Transgenic pyrimethamine-resistant plasmodium falciparum reveals transmission-blocking potency of P218, a novel antifolate candidate drug

Navaporn Posayapisit et al.

Summary: Antimalarial drugs targeting multiple parasite stages, especially the transmissible stages, are essential for malaria elimination. Developing new antifolate compounds to overcome antifolate resistance, including in mosquito development stages, is critical. The use of genome editing to create a pyrimethamine-resistant transgenic strain of Plasmodium falciparum provides a robust system for evaluating novel antifolate compounds.
