Glomerulosclerosis predicts poor renal outcome in patients with idiopathic membranous nephropathy

Cuimei Wei et al.

Summary: This study found that IMN patients in the Tertile3 group with a high proportion of glomerulosclerosis had more severe renal pathological damage, and glomerulosclerosis is a risk factor for renal function progression and poor renal prognosis in patients with IMN. As the proportion of glomerulosclerosis increases, the risk of renal endpoint events gradually increases.

Cooperative signaling between integrins and growth factor receptors in fibrosis

Horacio Maldonado et al.

Summary: Fibrosis is a pathological process characterized by fibrous connective tissue accumulation in organs, causing organ malfunction and failure. At the cellular level, aberrant fibroblast activation, ECM accumulation, and interaction between integrins and growth factor receptors play critical roles in fibrosis.

Retinoic acid receptor responder1 promotes development of glomerular diseases via the Nuclear Factor-κB signaling pathway

Katja Moller-Hackbarth et al.

Summary: Inflammatory pathways are activated in most glomerular diseases, with the induction of Rarres1 expression being a common molecular mechanism in chronic kidney diseases. The overexpression of Rarres1 in endothelial cells promotes kidney damage in a model of glomerulonephritis through the Nuclear Factor-kappa B signaling pathway, highlighting its pathogenic role in inflammation and fibrosis.

Targeting Interstitial Myofibroblast-Expressed Integrin αvβ3 Alleviates Renal Fibrosis

Jian Zhou et al.

Summary: Renal fibrosis is the end result of various chronic kidney diseases and current therapies do not effectively target interstitial myofibroblasts. PEGylated liposomes modified with cRGDfC show promise in specifically delivering drugs to these cells, leading to reduced fibroblast uptake and decreased proliferation and activation. The system has potential for renal fibrosis treatment with minimal side effects.

Pathophysiology of diabetic kidney disease: impact of SGLT2 inhibitors

Ralph A. DeFronzo et al.

Summary: Diabetic kidney disease is a leading cause of kidney failure globally, with over 50% of individuals entering dialysis or transplant programmes in the USA affected by it. Hyperglycaemia is the primary factor in its development, leading to various pathophysiological disturbances that contribute to progressive glomerular sclerosis and declining renal function. Multiple agents used in combination are likely to be needed to effectively slow the progression of the disease.

Summary: Metalloproteinases play a role in the development of aortic aneurysms and chronic kidney disease, damaging the aneurysm wall and renal tubular cells leading to fibrosis. Inhibitors of metalloproteinases have shown to reduce aneurysm growth and albuminuria, highlighting the importance of further research in improving prognosis through targeted treatments.

Renin-angiotensin system and inflammation update

Elena Cantero-Navarro et al.

Summary: The classical view of the renin-angiotensin system highlights its role in regulating sodium balance and blood pressure, but it is now recognized that RAS also plays a crucial role in organ damage and inflammation. Angiotensin II and its receptors can promote inflammatory responses, while other RAS components may have anti-inflammatory properties. Recent research has revealed new insights into the complex interactions between RAS and inflammation, shedding light on potential therapeutic targets for kidney and heart diseases.

Acute Kidney Injury is Aggravated in Aged Mice by the Exacerbation of Proinflammatory Processes

Laura Marquez-Exposito et al.

Summary: Aging increases the severity of acute kidney injury (AKI) following exposure to toxic compounds, potentially due to magnification of proinflammatory-related pathways in older mice. Older mice show a switch to proinflammatory cell death (necroptosis) instead of apoptosis, as well as overactivation of cellular senescence of resident renal cells and infiltrating inflammatory cells.
