S100 calcium-binding protein A9 from tumor-associated macrophage enhances cancer stem cell-like properties of hepatocellular carcinoma

Ran Wei et al.

Summary: The study revealed that TAMs can enhance the stem cell-like properties of HCC cells by upregulating the secreted protein S100A9, which in turn activates the NF-kB signaling pathway. Additionally, S100A9 can promote the recruitment of macrophages to HCC cells, suggesting a positive feedback loop between TAMs and HCC cells.

The New Immuno-Oncology-Based Therapies and Their Perspectives in Hepatocellular Carcinoma

Philippe Merle

Summary: Hepatocellular carcinoma is a poor prognosis tumor, with low response rates to immuno-oncology monotherapy. However, combination therapies involving immune checkpoint inhibitors and anti-angiogenic agents have shown breakthrough results in treating the disease.

Hepatocellular carcinoma in viral and autoimmune liver diseases: Role of CD4+CD25+Foxp3+regulatory T cells in the immune microenvironment

Alessandro Granito et al.

Summary: Over 90% of hepatocellular carcinoma cases occur in patients with cirrhosis, with hepatitis B virus and hepatitis C virus as leading causes. Immunotherapy has revolutionized HCC treatment by targeting immune cells rather than cancer cells, particularly focusing on regulatory T cells for their role in tumor progression and response to immune checkpoint inhibitors.

Aspects of the Tumor Microenvironment Involved in Immune Resistance and Drug Resistance

Khalil Khalaf et al.

Summary: The tumor microenvironment (TME) is a complex and dynamic tissue containing various components such as blood supply, immune cells, and extracellular matrix, which together create an environment conducive to tumor cell growth and proliferation. Cellular crosstalk, cell-to-ECM communication, exosomes, and specific metabolic patterns play crucial roles in immune evasion and therapy resistance in the TME. In addition, characteristics like hypoxia, metabolic abnormalities, and mechanical forces further contribute to treatment resistance.

Experience with regorafenib in the treatment of hepatocellular carcinoma

Alessandro Granito et al.

Summary: Regorafenib, structurally similar to sorafenib, shows significant survival advantage in unresectable hepatocellular carcinoma (HCC) patients who failed first-line treatment, by targeting angiogenic and tumor growth-promoting pathways, inhibiting tumor neo-angiogenesis and proliferation, and modulating the tumor microenvironment. It is being explored for use in combination therapy with immune checkpoint blockade and for extended use in patient categories beyond registrative studies.
