Butyrate in Energy Metabolism: There Is Still More to Learn

Lin Zhang et al.

Summary: Butyrate, as a main product of gut microbial fermentation, plays an important role in regulating whole body energy homeostasis. Research has shown that butyrate has multiple effects on metabolic health, including thermogenesis, lipid and glucose metabolism, appetite, inflammation, and influence on gut microbiota. Through pathways such as G protein-coupled receptors (GPRs) and acting as a histone deacetylase inhibitor, butyrate also impacts cellular systems.

An engineered genetic circuit for lactose intolerance alleviation

Mingyue Cheng et al.

Summary: The researchers developed a tri-stable-switch circuit with high beta-galactosidase activity and pH rescue ability, which can efficiently switch between enzyme expression in response to lactose and pH signals in bacterial cultures and mice. The circuit has the potential to alleviate lactose intolerance by stabilizing colon pH and maintaining beta-galactosidase activity, indicating its promising prospect as a prototype for the relief of lactose intolerance.

Gut microbes impact stroke severity via the trimethylamine N-oxide pathway

Weifei Zhu et al.

Summary: Clinical studies have shown associations between gut-microbiota-derived metabolite TMAO and stroke risk, but a causal role had not been proven. This study demonstrates that gut microbes impact stroke severity through choline and TMAO production, with traits being transmissible. The microbial cutC gene is identified as a key factor in TMA/TMAO production, cerebral infarct size, and functional impairment post-stroke.

Safety and pharmacodynamics of an engineered E. coli Nissle for the treatment of phenylketonuria: a first-in-human phase 1/2a study

Marja K. Puurunen et al.

Summary: The engineered E. coli strain SYNB1618 shows potential for the treatment of PKU, as it was safe and well tolerated in a phase 1/2a clinical trial with both healthy volunteers and PKU patients. The study demonstrated dose-responsive increases in strain-specific Phe metabolites in plasma and urine, supporting the proof of concept for using engineered bacteria in the treatment of rare metabolic disorders.
