Clinical, functional and genetic characterization of 16 patients suffering from chronic granulomatous disease variants - identification of 11 novel mutations inCYBB

Summary: Chronic granulomatous disease (CGD) is a rare inherited disorder characterized by a lack of NADPH oxidase activity in phagocytes. The X-linked form, X91-CGD, caused by mutations in the CYBB gene, is the most common. Through clinical, functional, and genetic characterizations of 16 cases, new mutations were identified and their pathological impacts were investigated. Low NADPH oxidase activity correlated with milder clinical forms of CGD, while X91(0)-CGD and X91(+)-CGD cases remained the most severe clinically.

Identification of Cytochrome b-245, beta-chain gene mutations, and clinical presentations in Iranian patients with X-linked chronic granulomatous disease

Summary: This study identified CYBB gene mutations in four Iranian patients with X-linked chronic granulomatous disease (X-CGD), including a novel insertion mutation. The patients exhibited different clinical features compared to other Iranian patients with the same mutations. The findings expand the genetic database of X-CGD patients from Iran and provide valuable information for carrier detection and prenatal diagnosis in high-risk families.

Summary: This study presents the clinical features, microbiological profile, and treatment challenges faced in managing liver abscesses in CGD patients at a tertiary care center in North-West India, highlighting the necessity for prolonged antimicrobial treatment and the potential benefits of glucocorticoids in reducing inflammation for early resolution of abscesses in CGD. Data from developing countries are limited in literature on this topic.

Clinical and Immunological Characteristics of 63 Patients with Chronic Granulomatous Disease: Hacettepe Experience

Summary: Through studying 63 patients with CGD, we found that in our sample from Turkey, the AR form of CGD was more frequent due to a high consanguinity rate, with gastrointestinal involvement found at relatively lower rates. The majority of patients received conventional prophylactic therapies, highlighting the outcomes of individuals who did not undergo HSCT.

Hemophagocytic Lymphohistiocytosis in Children with Chronic Granulomatous Disease-Single-Center Experience from North India

Summary: This study analyzed clinical and laboratory features of HLH in CGD patients in a tertiary-care center in North India. Among 80 patients diagnosed with CGD, 6.25% developed HLH, with infectious triggers including bloodstream infections, pneumonia, and splenic abscess. Notably, a child with X-linked CGD was infected with a fish pathogen, F. noatuensis. Early recognition and optimal management of both infectious trigger and HLH are important in preventing mortality.
