Acriflavine, a Potent Inhibitor of HIF-1α, Disturbs Glucose Metabolism and Suppresses ATF4-Protective Pathways in Melanoma under Non-hypoxic Conditions

Summary: The study found that HIF-1α is constitutively expressed in melanoma cells under normoxic conditions and its elevated expression is associated with the aggressiveness of melanoma tumors. Using acriflavine as an inhibitor of HIF-1α dimerization, the researchers investigated whether HIF-1α-regulated pathways contribute to the growth of melanoma cells under normoxia. Results showed that acriflavine can differentially modulate HIF-1α-regulated targets, inhibiting glucose metabolism and the Warburg effect, ultimately leading to cell death.

Summary: LINC00518 is a long noncoding RNA that is highly expressed in various cancers, including cutaneous malignant melanoma (CMM), and plays a role in promoting cancer progression. In CMM, LINC00518 induces radioresistance by regulating the glycolysis pathway through a negative feedback loop involving miR-33a-3p and hypoxia-inducible factor 1 alpha (HIF-1 alpha). Knockdown of LINC00518 expression sensitizes CMM cells to radiotherapy, suggesting a potential strategy to improve treatment outcomes.

Summary: The study found that ANXA3 promoted cell growth and migration in melanoma patients through activation of the HIF-1 alpha/VEGF signaling pathway. High ANXA3 expression was associated with lower overall survival and disease-free survival rates in patients. In contrast, down-regulation of ANXA3 reduced cell growth and migration and increased cytotoxicity in melanoma cells.
