Infliximab-based self-healing hydrogel composite scaffold enhances stem cell survival, engraftment, and function in rheumatoid arthritis treatment

Yue Zhao et al.

Summary: A new scaffold for rheumatoid arthritis management was developed through the integration of infliximab-based hydrogels and 3D printing porous metal scaffolds. The composite scaffold can modulate the survival and differentiation of ADSCs in vitro. In vivo results show that the composite scaffolds, along with ADSCs, are capable of suppressing inflammatory cytokines, rebuilding damaged cartilage, and improving subchondral bone repair.

Bioinspired stratified electrowritten fiber-reinforced hydrogel constructs with layer-specific induction capacity for functional osteochondral regeneration

Zhiguang Qiao et al.

Summary: This study developed a tri-layered scaffold mimicking the spatial variations of native osteochondral tissue to enhance osteochondral regeneration. By combining different materials and growth factors, mesenchymal stem cells were successfully induced to differentiate down both chondrogenic and osteogenic lineages, leading to simultaneous cartilage and subchondral bone regeneration. The inclusion of the S layer in the scaffold improved the regenerated cartilage's surface properties, showing promising applications for future use.

A hydrogel/particle composite with gradient in oxygen releasing microparticle for oxygenation of the cartilage-to-bone interface: Modeling and experimental viewpoints

Sajedeh Khorshidi et al.

Summary: The study developed a scaffold with a depth-dependent gradient of oxygen releasing microparticles for regeneration of cartilage-to-bone interface, showing potential to meet the different metabolic needs of cells. The mathematical modeling of flow velocity and microparticle concentration in the gradient making device, along with experimental evaluation of changes in morphology, chemistry, and structure of the hydrogel/particle composite, provided insights into the complexity and efficacy of the designed scaffold.

High-precision, gelatin-based, hybrid, bilayer scaffolds using melt electro-writing to repair cartilage injury

Yu Han et al.

Summary: Articular cartilage injury is a common disease in orthopedics due to poor self-repairing ability. This study developed a novel printing method using MEW technology to prepare tissue engineering scaffolds with high porosity and high precision. By mixing gelatin with poly (lactic-co-glycolic acid) to create high-concentration printer ink, a composite scaffold with bioactive properties was constructed to promote cartilage repair using autologous bone marrow mesenchymal stem cells.
