Summary: The effects of cyclophosphamide on anagen hair follicles depend on dosage, with low dosages leading to dystrophy that can be self-repaired, while high dosages prematurely induce other growth stages. Analysis of cell dynamics revealed that apoptosis of central cells in hair bulbs is repairable, bulge stem cells remain unaffected, and basal cup cells can quickly replenish damaged cells and reconstruct structures.

Inhibition of Shh Signaling through MAPK Activation Controls Chemotherapy-Induced Alopecia

Summary: Chemotherapy-induced hair loss (CIA) is a major issue in clinical oncology. Research has shown that downregulation of the Shh signaling pathway in hair follicles is a critical early event in CIA. Inhibition of Shh signaling leads to hair loss, while activation of the MAPK pathway is a key upstream event that can be manipulated to rescue CIA. Targeting the intrafollicular MAPK-Shh axis may provide a promising strategy for managing CIA.
