TMBIM6 (transmembrane BAX inhibitor motif containing 6) enhances autophagy through regulation of lysosomal calcium

Summary: TMBIM6 enhances autophagy by increasing lysosomal Ca2+ release and subsequently activating TFEB, leading to enhanced lysosome biogenesis to alleviate metabolism stress. Genetic inactivation of TMBIM6 reduces lysosomal Ca2+ and TFEB nuclear translocation, resulting in increased autophagy flux. TMBIM6, an ER membrane protein, is suggested to be a lysosomal Ca2+ modulator that coordinates with autophagy to alleviate metabolism stress.
