Macrophage targeting in cancer

Martha Lopez-Yrigoyen et al.

Summary: Tumorigenesis is influenced by interactions between cancer cells and the tumor microenvironment. Tumor-associated macrophages play a crucial role in tumor development, with the potential to both promote antitumor responses and exhibit immunosuppressive protumor phenotypes. Targeting TAMs has become a strategy in cancer therapy, with a focus on reprogramming TAMs to support antitumor immune responses.

c-Myc promotes lymphatic metastasis of pancreatic neuroendocrine tumor through VEGFC upregulation

Tsung-Ming Chang et al.

Summary: Pancreatic neuroendocrine tumor (pNET) is a type of pancreatic tumor with neuroendocrine differentiation. Early stage pNET can be treated with surgical resection for long-term survival, while pNET with metastasis has a poor prognosis. In this study, it was found that c-Myc overexpression is associated with lymph node metastasis in pNET xenograft mice. Combination therapy using an mTOR inhibitor and c-Myc inhibitor or VEGFC-neutralizing protein reduced lymph node metastasis in mice with c-Myc overexpression, indicating potential therapeutic targets for pNET.

Breaking the cycle: Targeting of NDRG1 to inhibit bi-directional oncogenic cross-talk between pancreatic cancer and stroma

Bekesho Geleta et al.

Summary: The study reveals that NDRG1 plays an inhibitory role in the interaction between PaCa and PSCs, reducing signaling pathways like HGF and IGF-1 and inhibiting metastasis and cell migration in pancreatic cancer. DpC, a novel anticancer agent, upregulates NDRG1 expression to decrease PaCa cells' sensitivity to ligands secreted by PSCs.

Fat1 deletion promotes hybrid EMT state, tumour stemness and metastasis

Ievgenia Pastushenko et al.

Summary: This study reveals that mutation of the FAT1 gene promotes tumor initiation, progression, invasiveness, stemness, and metastasis. Loss of function of FAT1 activates a CAMK2-CD44-SRC axis while inactivates EZH2, playing important roles in the expression of YAP1 and ZEB1.

Targeting metastatic cancer

Karuna Ganesh et al.

Summary: Improved understanding of metastases biology has led to advancements in targeting and treating both micro- and macrometastases. Recent studies have identified the unique biology of metastasis-initiating cells, providing new opportunities for developing more effective treatment strategies through cancer sequencing, mechanistic studies, and clinical trials. These insights offer potential for better outcomes in targeting metastatic relapse and improving patient survival.

A novel HDGF-ALCAM axis promotes the metastasis of Ewing sarcoma via regulating the GTPases signaling pathway

Yang Yang et al.

Summary: Research has shown that HDGF promotes metastasis in Ewing sarcoma (ES) and leads to poor patient outcomes by regulating multiple metastasis-associated genes, particularly ALCAM. The study further reveals associations among HDGF, ALCAM, and GTPases, suggesting that the HDGF/ALCAM/GTPases axis may serve as a promising therapeutic target for limiting ES metastasis.

VEGF-C mediates tumor growth and metastasis through promoting EMT-epithelial breast cancer cell crosstalk

Deguang Kong et al.

Summary: VEGF-C secreted by EMT breast cancer cells can promote proliferation, migration, and invasion of epithelial breast cancer cells through non-canonical activation of GLI signaling. Inhibiting VEGF-C in EMT cells or knocking down NRP2 in neighboring epithelial cancer cells disrupts the aggressive phenotypes induced by EMT cells, suggesting a novel therapeutic target for metastasis in breast cancer.

Co-Overexpression of TWIST1-CSF1 Is a Common Event in Metastatic Oral Cancer and Drives Biologically Aggressive Phenotype

Sabrina Daniela da Silva et al.

Summary: There is clinical evidence linking ulcerated and inflammatory cell-infiltrated oral cancer with early metastases. Genomic screening in patients with metastatic oral cancer revealed specific gene changes related to macrophage chemotaxis and tumor progression. Overexpression of TWIST1 and CSF1 was observed in metastatic cases, leading to poor patient survival, and further studies showed their cooperation in promoting oral squamous cell carcinoma invasiveness.

Activation of WNT7b autocrine eases metastasis of colorectal cancer via epithelial to mesenchymal transition and predicts poor prognosis

Shuai Jiang et al.

Summary: This study demonstrated that activation of WNT7b autocrine may contribute to colorectal cancer metastasis by triggering the EMT process through the Wnt/β-catenin signaling pathway. High levels of WNT7b autocrine secretion predict a poor outcome in patients with colorectal cancer and may be a promising candidate for clinical treatments.

Wnt/β-catenin/Slug pathway contributes to tumor invasion and lymph node metastasis in head and neck squamous cell carcinoma

Jung Hwa Moon et al.

Summary: The canonical Wnt/β-catenin/Slug signaling axis plays a significant role in cancer cell invasion and lymph node metastasis in head and neck squamous cell carcinoma (HNSCC), and is a major predictive factor for patient survival rate. Blocking this pathway may be a potential treatment strategy for LNM and tumor recurrence in HNSCC.

TGF-β-induced DACT1 biomolecular condensates repress Wnt signalling to promote bone metastasis

Mark Esposito et al.

Summary: Intracellular signaling involves a diverse array of molecular players and spatial segregation mechanisms, with liquid-liquid phase separation identified as a distinct mechanism. DACT1 induced by TGF-beta forms protein condensates in the cytoplasm to repress Wnt signaling, impacting breast and prostate cancer bone metastasis through its role in sequestering CK2.

Cancer-associated fibroblast secretion of PDGFC promotes gastrointestinal stromal tumor growth and metastasis

Hyunho Yoon et al.

Summary: This study identified a paracrine signaling network in the tumor microenvironment where cancer-associated fibroblasts drive GIST growth and metastasis through the production of PDGFC. Targeting CAFs with a dual PI3K/mTOR inhibitor synergized with imatinib to increase tumor cell killing. This new cellular target for GIST therapy may improve disease control and cure rates.

Stromal NRG1 in luminal breast cancer defines pro-fibrotic and migratory cancer-associated fibroblasts

Mireia Berdiel-Acer et al.

Summary: High expression of HER3 in luminal breast cancer subtypes is regulated by NRG1, which is secreted by cancer-associated fibroblasts (CAFs). The study also identifies a HER3-independent NRG1 signaling pathway in CAFs that induces a migratory and pro-fibrotic phenotype. Additionally, Hyaluronan Synthase 2 (HAS2) is identified as a key player supporting NRG1 signaling in CAFs.
