Circulating ceramides as biomarkers of cardiovascular disease: Evidence from phenotypic and genomic studies

Heritability and family-based GWAS analyses of the N-acyl ethanolamine and ceramide plasma lipidome

Summary: Signalling lipids of the N-acyl ethanolamine (NAE) and ceramide (CER) classes have emerged as potential biomarkers of cardiovascular disease (CVD) in genetic studies, showing heritability and influence from common DNA variants. However, Mendelian randomization studies did not reveal a significant association between genetically determined differences in CER levels and CVD or diabetes. Novel genetic loci like SGPP1 and DEGS1 suggest a causal association between CERs and hematological phenotypes, based on 2-sample Mendelian randomization analyses in large cohorts like the UK Biobank and INTERVAL.
