Genome mining combined metabolic shunting and OSMAC strategy of an endophytic fungus leads to the production of diverse natural products

Summary: Endophytic fungi, such as Penicillium dangeardii, have the potential to produce bioactive small molecules, but some biosynthetic gene clusters may be silent. By employing a metabolic shunting strategy and optimizing culture conditions, multiple silent genes encoding for other polyketide synthases were activated in P. dangeardii, leading to the identification of a variety of new compounds.

Sustainable production of camptothecin from an Alternaria sp. isolated from Nothapodytes nimmoniana

Summary: The research aims to identify a sustainable and high-yielding camptothecin producing endophyte as an alternative source for commercial production of camptothecin. A total of 132 endophytic fungal strains were isolated from different parts of the Nothapodytes nimmoniana plant, out of which 94 were found to produce camptothecin in suspension culture.

Summary: This study used a metagenomics approach to investigate the endophytic microbial diversity associated with Piper longum, revealing significant differences in diversity and abundance of bacterial and fungal endophytes between spikes and leaves. Bacterial genera in spikes were reported to produce bioactive compounds, while different bacterial genera were predominant in leaves. Fungal endophytes in spikes were found to produce commercially important metabolites, indicating their potential for bioprospecting.

Summary: Under natural conditions, plants are not individuals but are associated with a diverse microbiota to form the plant holobiont. Endophytes, a type of plant-associated microbes residing within plant tissues, have shown various modes of symbiotic association with plants, from beneficial to pathogenic. The factors influencing plant-endophyte interactions and the mechanisms allowing endophytes to colonize plants without triggering a strong defense reaction are still being explored.

Identification of a Novel Biosynthetic Gene Cluster in Aspergillus niger Using Comparative Genomics

Summary: The study found that a biosynthetic gene cluster in Aspergillus niger is downregulated in co-culture with Bacillus subtilis. Gene analysis revealed that the cluster contains various genes including nonribosomal peptides synthetase and transcription factor. Overexpression of the transcription factor gene led to reduced growth rate and production of two previously unknown compounds, which were reverted to wild-type phenotype upon deletion of the nonribosomal peptides synthetase gene. This suggests that the biosynthetic gene cluster is regulated by the transcriptional regulator gene and is involved in the production of these compounds.

Evaluation of Apple Root-Associated Endophytic Streptomyces pulveraceus Strain ES16 by an OSMAC-Assisted Metabolomics Approach

Summary: The OSMAC approach was used to culture the endophytic Streptomyces pulveraceus strain ES16 under various conditions, leading to the production of geosmin, polycyclic tetramate macrolactams (PTMs), and mirubactin, as revealed by SPME-GC-MS, HPLC-HRMSn, and MALDI-HRMSI. These compounds were found to play important roles in attracting soil arthropods, providing chemical defense against pathogens, and promoting ecological interactions between the endophyte and its host plant.
