The use of negative control outcomes in Mendelian randomization to detect potential population stratification

Eleanor Sanderson et al.

Summary: Mendelian randomization analysis assumes no association between genetic variants and outcomes other than through the exposure of interest, but population stratification can introduce confounding. Negative control outcomes are suggested for detecting potential confounding in MR studies. Regular use of negative control outcomes is recommended to detect population stratification in MR studies.

Effect of selection bias on two sample summary data based Mendelian randomization

Kai Wang et al.

Summary: Mendelian randomization (MR) is increasingly used to infer causal relationships between exposure and traits, but traditional methods may suffer from selection bias and invalidate MR findings. A simpler and more powerful alternative has been proposed.
