Spatiotemporally programmable cascade hybridization of hairpin DNA in polymeric nanoframework for precise siRNA delivery

Feng Li et al.

Summary: DNA nanostructures have been used as carriers for gene delivery, but spatiotemporally programmable assembly of DNA under nanoconfinement is challenging due to DNA complexity. A strategy using cascade hybridization chain reaction of DNA hairpins in polymeric nanoframework enables precise siRNA delivery. The nanoframework provides stability and ease of functionalization, addressing DNA complexity issues. Phenylboronate installation on nanoframework enhances cellular uptake and lysosomal escape.

Protein-Based Hybrid Responsive Microparticles for Wound Healing

Hui Zhang et al.

Summary: The development of controllable responsive microparticles composed of natural polymer materials, loaded with growth factors and antibacterial peptides, shows promising potential for drug release and wound healing applications. Under near-infrared (NIR) irradiation, these microparticles can achieve desired controllable release of growth factors to promote neovascularization formation. Additionally, the antibacterial peptides embedded in the microparticles provide antibacterial ability during storage and usage. These characteristics make the microparticles ideal for drug delivery and wound healing.

An injectable and biodegradable nano-photothermal DNA hydrogel enhances penetration and efficacy of tumor therapy

Liping Zhou et al.

Summary: Researchers developed highly permeable, photothermal, injectable, and positively charged biodegradable nucleic acid hydrogel (DNA-gel) nanoparticles for delivering cancer drugs. These nanoparticles, derived from photothermal materials containing black phosphorus quantum dots (BPQDs), enhanced tumor cell sensitivity to photothermal and photodynamic therapy. In mouse models, the DNA gel nanoparticles showed effective anti-tumor activity by clearing both small and large tumor masses, reducing drug resistance, and improving overall survival rates in mice with breast tumors.

Activating an adaptive immune response from a hydrogel scaffold imparts regenerative wound healing

Donald R. Griffin et al.

Summary: The study aimed to slow MAP scaffold degradation by changing the chirality of peptides, but unexpectedly, D-peptide crosslinked MAP scaffolds led to accelerated degradation in vivo, promoting tissue regeneration and enhancing wound healing. D-MAP triggered an immune response against D-chiral peptides and intact adaptive immune system was crucial for skin regeneration induced by the hydrogel.
