A Nanoplasmonic Assay of Oligonucleotide-Cargo Delivery from Cationic Lipoplexes

Summary: A powerful new biophysical model is used to study the permeability of nanocarrier lipid membranes and the release mechanism of oligonucleotides. Through the combined use of surface enhanced Raman spectroscopy and fluorescence lifetime correlation spectroscopy, a novel mechanism for oligonucleotide release facilitated by lipoplex-mediated endosomal-escape is identified, indicating a complex multistep interplay between cargo and target membrane. This approach presents a new paradigm for assessing vector delivery and offers a potential solution to engineering the release mechanism of large therapeutic molecules from nanocarriers.
