The current drug discovery landscape for trypanosomiasis and leishmaniasis: Challenges and strategies to identify drug targets

Summary: Human trypanosomiasis and leishmaniasis, caused by protozoan parasites, have become globally distributed tropical diseases due to human migration, climate change, and anthropogenic disturbance. The current chemotherapy for these diseases has limited efficacy and drug resistance is a growing concern. This review discusses the limitations of available drugs and highlights promising leads from current drug-based clinical trials. It also examines the challenges in target-based drug discovery and the advantages and limitations of modern research tools.

Epigallocatechin-3-gallate exhibits antiviral effects against the duck Tembusu virus via blocking virus entry and upregulating type I interferons

Summary: This study demonstrated that EGCG can inhibit DTMUV infection by reducing virus titers, virus copy number, and viral E protein expression, as well as direct virucidal abilities and inhibition of virus adsorption. EGCG also suppressed DTMUV replication in cells by upregulating interferon alfa and interferon beta levels, ultimately improving survival rates in infected ducklings.

Poplar-type propolis is the most widespread in the world, mainly sourced from poplar trees but with contributions from other plants as well. The aim of research is to identify the chemical composition and activities of plant resins and balms used by bees to produce propolis and compare them with similar products from other geographical regions.

Clinical trials are needed to investigate its effects on individuals with viral diseases, either alone or in combination with antiviral drugs or vaccines. Specific research on the effects of propolis on COVID-19, either in vitro or in infected individuals, should also be conducted to explore its potential as a treatment option.

Summary: Bee honey and propolis are rich in bioactive compounds with antimicrobial, antiviral, and anti-inflammatory properties, showing potential benefits in fighting COVID-19. Flavonoids in propolis and honey may inhibit viral fusion, interactions that trigger cytokine storm, and viral replication. Propolis extracts delivered by nanocarriers exhibit better antiviral effects against SARS-CoV-2 than ethanolic extracts, showing promise as a potential adjuvant treatment for COVID-19.
