Summary: In all branches of life, stalled translation intermediates are recognized and processed by ribosome-associated quality control pathways. The process involves splitting of stalled ribosomes and targeting incomplete proteins for degradation by NEMF family proteins. Structural analysis reveals the mechanism of C-terminal tailing in Bacillus subtilis, mediated by NEMF protein RqcH and RqcP, in synthesizing polypeptides in the absence of mRNA or the small subunit.

Heat activates the AAA plus HslUV protease by melting an axial autoinhibitory plug

Summary: At low temperatures, protein degradation by the AAA+ HslUV protease is slow due to residues in the intermediate domain of the HslU(6) unfoldase blocking the axial channel, preventing substrate binding and degradation until heat-induced melting of this autoinhibitory plug activates HslUV proteolysis through a model supported by biochemical experiments with wild-type and mutant enzymes.

Harnessing Machine Learning To Unravel Protein Degradation in Escherichia coli

Summary: The degradation of intracellular proteins in Gram-negative bacteria is regulated through various cellular processes and serves as a mechanism for quality control. By employing a quantitative pulsed-SILAC method followed by mass spectrometry analysis, this study identified fast-degrading novel proteins and developed a machine learning classifier to discriminate them from the rest of the proteome. These findings expand the understanding of protein degradation in bacteria and have implications for protein engineering, potentially aiding in the identification of new antibacterial drug targets.
