Evaluating Mismatch Repair/Microsatellite Instability Status Using Cytology Effusion Specimens to Determine Eligibility for Immunotherapy

Elizabeth M. Jacobi et al.

Summary: This study investigated the feasibility of using cell block preparations for MMR IPOX evaluation and found a high concordance of MMR IPOX testing between cytologic effusion specimens and surgical pathology specimens. However, some cases exhibited limitations such as low tumor cell numbers and unclear staining in individual cells.

Identifying mismatch repair-deficient colon cancer: near-perfect concordance between immunohistochemistry and microsatellite instability testing in a large, population-based series

Maurice B. Loughrey et al.

Summary: This study compared PCR-based microsatellite instability (MSI) testing and MMR protein immunohistochemistry (IHC) in colorectal cancer cases and found that both methods are equally proficient in establishing MMR/MSI status, but awareness of potential pitfalls is important. The choice of methodology may depend on available services and expertise.

Detection of microsatellite instability in a panel of solid tumours with the Idylla MSI Test using extracted DNA

Adrien Pecriaux et al.

Summary: The aim of the study was to evaluate the Idylla MSI Test by Biocartis for MSI analysis, which can be completed within 150 minutes. The results showed high sensitivity (94%) and specificity (100%), with a few invalid and false-negative results observed. The experiment also emphasized the importance of DNA quantity loaded into the cartridge, with a LOD between 4% and 8%.

Idylla microsatellite instability assay versus mismatch repair immunohistochemistry: a retrospective comparison in gastric adenocarcinoma

Luke Farmkiss et al.

Summary: Up to 22% of gastric adenocarcinomas belong to the microsatellite instability (MSI) subtype, which responds to novel immunotherapies but may not benefit from conventional chemotherapy. Screening for MSI subtype in gastric adenocarcinoma can help detect inherited diseases and identify at-risk relatives. MSI testing and mismatch repair (MMR) immunohistochemistry (IHC) are established methods for detecting this subtype, with the best results achieved through their combined use.

MSIsensor-ct: microsatellite instability detection using cfDNA sequencing data

Xinyin Han et al.

Summary: MSIsensor-ct is a bioinformatics tool based on a machine learning protocol for detecting MSI status using cfDNA sequencing data, with a potential stable MSIscore threshold of 20%. Evaluation on independent testing datasets with various levels of ctDNA and sequencing depth showed 100% accuracy within the limit of detection of 0.05% ctDNA content. MSIsensor-ct requires only BAM files as input, making it user-friendly and easily integrated into NGS analysis pipelines.

NGS-based identification and tracing of microsatellite instability from minute amounts DNA using inter-Alu-PCR

Fangyan Yu et al.

Summary: Sensitive detection of microsatellite instability (MSI) using next generation sequencing (NGS) is increasingly important in cancer prognosis and prediction. A new approach combining inter-Alu-PCR with targeted NGS offers a practical and efficient method for detecting MSI, with the potential to trace low-level MSI in tissue or liquid biopsies.

Summary: Precision medicine is crucial for designing personalized treatment strategies to extend patient survival. Cell-free tumor DNA (ctDNA) is a noninvasive biomarker that shows promise in selecting patients for genomic-driven therapies. Standardization of ctDNA collection and analysis methods is essential for its wide adoption in clinical practice.
