Tissue-specific transcriptional imprinting and heterogeneity in human innate lymphoid cells revealed by full-length single-cell RNA-sequencing

Luca Mazzurana et al.

Summary: This study utilized single-cell RNA sequencing to uncover tissue-specific transcriptional profiles and heterogeneity of CD127(+) ILCs in four human tissues. It identified migratory properties, tissue-residency, activation, and modified metabolism characteristics of ILCs in different tissues. The study also revealed potential differentiation pathways from naive to mature ILC subsets, and unique TCR-V(D)J-rearrangement patterns of blood ILC1-like cells.

Single-cell insights into the hematopoietic generation of T-lymphocyte precursors in mouse and human

Ellen V. Rothenberg

Summary: Recent studies have revealed a robustly conserved core program in early T-cell precursors in mice and humans, with some informative variations between species and ontogenic states. The T-cell program is closely related to dendritic cell programs, especially plasmacytoid dendritic cells in humans.

RORα is a critical checkpoint for T cell and ILC2 commitment in the embryonic thymus

Ana C. F. Ferreira et al.

Summary: ILC2 cells can develop in the embryonic thymus from shared T cell precursors before the emergence of double-positive T cells, and migrate to mucosal tissues. The expression of ROR alpha in the thymus promotes ILC2 development while repressing T cell development. Notch signaling, BCL11B, and ROR alpha play critical roles in the co-development of T cells and ILC2 cells in the thymus.

Group 2 innate lymphoid cells support hematopoietic recovery under stress conditions

Takao Sudo et al.

Summary: This study reveals that BM-resident ILC2s support the recovery of HSPCs by secreting GM-CSF and interact with B cells to maintain hematopoietic homeostasis. ILC2s function by "sensing" damaged BM spaces and supporting hematopoietic recovery under stress conditions.

Liver type 1 innate lymphoid cells develop locally via an interferon-γ-dependent loop

Lu Bai et al.

Summary: Research indicates that the adult mouse liver contains lineage-CD122(+)CD49a(+) progenitor cells for liver ILC1s derived from fetal liver. The production of IFN-gamma by liver ILC1s themselves promotes their development in situ, emphasizing the role of extramedullary hematopoiesis in liver immune composition.

How transcription factors drive choice of the T cell fate

Hiroyuki Hosokawa et al.

Summary: Transcription factors play essential roles in early thymic development to establish T cell lineage identity by controlling the activities of specific genomic loci. The interactions between inherited epigenetic states, transcription factor-DNA binding affinity thresholds, and influences of given transcription factors on other factors in the same cells determine T cell identity and make the lineage choice irreversible.
