Real-time dynamics of Plasmodium NDC80 reveals unusual modes of chromosome segregation during parasite proliferation

Mohammad Zeeshan et al.

Summary: Studies have revealed that Plasmodium displays unusual nuclear division characteristics during cell proliferation, with highly abnormal kinetochore arrangements observed during the reproductive stage. By studying the behavior and composition of the NDC80 complex, a divergent candidate for the SPC24 subunit in Plasmodium has been identified, completing a canonical, albeit unusual, NDC80 complex structure. In summary, the kinetochore has been shown to be an ideal tool for investigating the non-canonical modes of chromosome segregation and cell division in Plasmodium.

Dynamic Chromatin Structure and Epigenetics Control the Fate of Malaria Parasites

Thomas Hollin et al.

Summary: The human malaria parasite, Plasmodium falciparum, has developed complex molecular mechanisms to ensure survival, spread, and transmission to its next host. Evidence suggests that the parasite has complex and complementary layers of regulatory mechanisms controlling gene expression, including epigenetics, transcription factors, and nuclear architecture, to control gene regulation and life cycle progression.

Internal daughter formation of Toxoplasma gondii tachyzoites is coordinated by transcription factor TgAP2IX-5

Chaoyue Wang et al.

Summary: This study evaluated the effect of the transcription factor TgAP2IX-5 on daughter cell formation in T. gondii, revealing that the loss of TgAP2IX-5 leads to defects in internal daughter IMC formation and abnormalities in organelle morphology during cell division.

Depletion of the mini-chromosome maintenance complex binding protein allows the progression of cytokinesis despite abnormal karyokinesis during the asexual development of Plasmodium falciparum

Sabrina Absalon et al.

Summary: This study identifies a potential cell cycle checkpoint and key protein in malaria parasites, which is involved in maintaining nuclear morphology and parasite proliferation. Depletion of PfMCMBP leads to the production of abnormal merozoites, indicating that the parasite lacks a mechanism to prevent abnormal cytokinesis.

TgAP2IX-5 is a key transcriptional regulator of the asexual cell cycle division in Toxoplasma gondii

Asma S. Khelifa et al.

Summary: Khelifa et al. identified a key transcriptional regulator, TgAP2IX-5, in the model Apicomplexan Toxoplasma gondii, which controls the timing of daughter cell emergence and regulates the expression of necessary transcription factors for completion of the budding cycle. Additionally, TgAP2IX-5 acts as a limiting factor by promoting the inhibition of the differentiation pathway, ensuring the continuation of asexual proliferation.
