Summary: De novo duplications in the ATAD3 gene locus cause distinctive mitochondrial disease symptoms such as lethal perinatal cardiomyopathy. These duplications can be identified through genome sequencing but are often missed by microarray sequencing. The ATAD3 duplications result in a significant reduction in mitochondrial oxidative phosphorylation complex I and its activity.
