Hallmarks of Health

Carlos Lopez-Otin et al.

Summary: Health is more than just the absence of disease; it involves a combination of organizational and dynamic features that maintain physiology. Biological causes of health include spatial compartmentalization, maintenance of homeostasis, and adequate responses to stress. Disruption of these features can lead to pathogenic processes.

Apaf-1 Pyroptosome Senses Mitochondrial Permeability Transition

Wanfeng Xu et al.

Summary: The study identified the Apaf-1 pyroptosome as a crucial machinery for cells sensing MPT signals and shedding light on how cells execute intrinsic pyroptosis under sterile conditions, showing that Caspase-4 and GSDME-induced pyroptosis play a critical role in cholestatic liver failure.

NINJ1 mediates plasma membrane rupture during lytic cell death

Nobuhiko Kayagaki et al.

Summary: Plasma membrane rupture is a cataclysmic event in lytic cell death, releasing damage-associated molecular patterns to propagate the inflammatory response. NINJ1 protein plays an essential role in inducing PMR, with Ninj1-deficient macrophages showing impaired release of intracellular proteins and distinctive ballooned morphology. This study overturns the long-held belief that cell death-related PMR is a passive event, highlighting the active role of NINJ1 in mediating PMR.

Inflammasome-Dependent Coagulation Activation in Sepsis

Runliu Wu et al.

Summary: Sepsis is a potentially life-threatening condition caused by a dysregulated host response to infection, leading to systemic inflammation and coagulation activation, ultimately resulting in multiple organ failure. Recent studies have highlighted the important role of inflammasomes in immunocoagulation, which are regulated by various factors.
