Enantioselective Alkylamination of Unactivated Alkenes under Copper Catalysis

Summary: An enantioselective addition reaction of various alkyl groups to unactivated internal alkenes under Cu catalysis has been developed, leading to high yields of β-lactams featuring two contiguous stereocenters with excellent enantioselectivity. Mechanistic studies suggest that a nucleophilic addition of the alkyl radical to Cu-II-coordinated alkene is the enantio-determining step.

Enantioselective α-Arylation of Ketones via a Novel Cu(I)-Bis(phosphine) Dioxide Catalytic System

Summary: A novel catalytic system based on copper(I) and chiral bis(phosphine) dioxides has been developed for the arylation of silyl enol ethers, yielding enolizable alpha-arylated ketones with high yields and enantiomeric excess up to 95%. This method is applicable to noncyclic ketones and complements other palladium-catalyzed approaches. Optimization of ligand structure was achieved through rational design driven by correlation analysis, and preliminary mechanistic hypotheses were evaluated to understand the role of chiral bis(phosphine) dioxides.

Summary: This study presents a novel approach to achieve stereoselective synthesis of highly functionalized 1,3-diene products using a stereoselective Ni-catalyzed alkyne vinylation process without the need for exogenous base. Experimental and theoretical studies support the proposed mechanism involving generation of Ni(0) species in situ and subsequent oxidative cyclization with B-assisted C-O bond cleavage. The highly functionalized products have the potential to serve as important building blocks for synthesizing complex organic architectures.
